Repositori BKPKKarakteristik Protein Antigen M.tuberculosis Multi Resisten Obat Anti Tuberculosis dengan Cara Imunobloting Menggunakan Serum Pasien TBSarwoHandayaniauthorKasus resistensi kuman M.tuberculosis terhadap Obat Anti Tuberkulosis (OAT) terus meningkat. Resistensi tersebut berhubungan dengan terjadinya mutasi titik yang berpengaruh terhadap kerja beberapa enzim dalam mensintesa protein kuman. Telah dilakukan penelitian untuk mengetahui karakterisasi protein antigen kuman M.tuberculosis yang resisten terhadap OAT dengan cara membandingkannya dengan protein antigen dari kuman yang sensitif terhadap OAT. Karakteristik protein antigen dilakukan dengan cara imunobloting menggunakkan tidak adanya perbedaan antara kedua protein antigen kuman tersebut, karena hanya terlihat satu pita protein yang dengan berat molekul 70 kd. Hal tersebut kemungkinan disebabkan oleh karena protein belum terpecah dengan baik, sehingga hanya terlihat satu pita protein saja, kadar protein yang diperoleh rendah, isolasi protein belum sempurna atau adanya protein yang rusak selama proses ekstraksi sehingga protein tersebut tidak tampak. Untuk mendapatkan hasil protein yang lebih baik perlu dipertimbangkan beberapa hal diantaranya : proses pembiakan kuman, protein yang akan diisolasi, sifat protein yang akan diisolasi dan metode yang digunakan untuk isolasi protein.
