Pemeriksaan dilakukan dengan tes ELISA menggunakan antigen Plasmodium falciparum strain Manado. Dari 62 sera penderita malaria falciparum yang diperiksa hanya 35 (56,45%) yang menunjukkan hasil positif. Dari 88 serta penderita malaria vivax dan 6 sera penderita infeksi campuran (Pf + Pv) didapati 26 (29,55%) dan 4 (66,67%) sera positif. Pada penderita malaria falciparum terdapat hubungan antara nilai OD (Optimal Density) dengan kepadatan parasit dimana semakin tinggi nilai OD semakin kecil kepadatan parasit. Disamping itu semakin tinggi nilai OD semakin kecil waktu yang dibutuhkan untuk bebas panas dan bebas parasit. Pada penderita juga terlihat terjadi peningkatan angka sero positif dari 50,85% pada saat sebelum pengobatan menjadi 67,8% selama pengobatan dan turun kembali menjadi 61,02% pada hari ke-28 setelah pengobatan.
