Carra pemeriksaan ini kurang sensitif dan memerlukan waktu yang relatif lama. Hal ini sangat berpengaruh pada penentuan pengobatan dan usaha memutus rantai penularan. Penggunaan PCR (Polymerase Chain Reaction) yaitu suatu metode memperbanyak (amplifikasi) DNA invitro secara enzimatik telah dikembangkan untuk mendeteksi M. tuberculosis, teknik ini diketahui mempunyai sensitivitas dan spesifitas yang tinggi. Hasil yang didapat adalah pemeriksaan sputum positif mikrospkopis (BTA), positif biakan 28 sampai 26 sampel diantaranya menunjukkan positif pada tes PCR. Pemeriksaan dengan hasil negatif mikroskopis (BTA), positif biakan 12 sampel, 11 sampel diantaranya menunjukkan hasil positif pada tes PCR. Dari 60 sampel yang menunjukkan negatif mikroskopis dan negatif biakan, pada tes PCR menunjukkan 19 sampel positif. Secara keseluruhan didapat hasil, dari 100 sputum yang diperiksa 31% positif secara mikroskopis, 38% positif dengan biakan dan 56% positif dengan tes PCR. Dari hasil evaluasi, penggunaan PCR untuk diagnosis tuberkulosis paru ternyata lebih sensitif dan lebih cepat dibandingkan dengan pemeriksaan bakteriologik secara mikroskopik dan biakan.
