Tujuan penelitian ini adalah mendeteksi R.typhi pada pinjal tikus dari tiga daerah pelabuhan yaitu Semarang, Kupang, dan Maumere. Deteksi R.typhi dilakukan dengan amplifikasi gen gltA (primer 877F dan 1258R) serta terhadap gen rompB secara nested. Sekuensing dilakukan untuk mengkonfirmasi spesies R.typhi. Hasil amplifikasi gen gltA diperoleh persentase Rickettsia spp masing-masing 19%, 61%, dan 44% di Semarang, Kupang, dan Maumere. Hasil amplifikasi gen rompB memberikan 6 hasil positif, 4sampel dari Kupang merupakan Spotted Fever Group (SFG) dan 2 sampel dari Semarang merupakan Typhus Group (TG). Delapan belas sampel disekuensing terkonfirmasi sebagai R.typhi pada 7 sampel, dan 11 lainnnya merupakan Bartonella sp.
