Kemudian, dilanjutkan dengan pemeriksaan fisik dan dipertegas dengan pemeriksaan laboratorium. Pemeriksaan laboratorium mempunyai peranan yang penting dalam diagnosis penderita demam berdarah dengue. Namun, diagnosis pasti belumlah dapat ditegakkan mengingat tanda patognomonik baru dapat ditentukan dengan ditemukannya virus dengue pada penderita demam berdarah. Namun, pemeriksaan laboratorium lain dengan akurasi dan presisi tinggi dapat menunjang diagnosis penyakit. (Sumarmo SPS, 1983). Pemeriksaan laboratorium standar yang diajukan oleh WHO (1997) yang diadaptasi oleh Departemen Kesehatan Republik Indonesia untuk DBD adalah Trombositopeni dan Uji torniquet (Thomas Suroso dkk, 2000).
